Andembry (garadacimab-gxii) is an activated Factor XII (FXIIa) inhibitor (monoclonal antibody) used for prophylaxis to prevent attacks of hereditary angioedema (HAE) in adult and pediatric patients aged 12 years and older.

Recommended Dosage:
The recommended dosage of Garadacimab-gxii is an initial loading dose of 400 mg (two injections of 200 mg) administered subcutaneously on the first day of treatment, followed by a maintenance dosage of 200 mg administered subcutaneously every month.
In case any dose of Garadacimab-gxii is missed, administer the dose as quickly as possible.

Warning & Precautions

  • Treatment with Garadacimab‑gxii 200 mg/1.2 mL may be responsible for interfering with activated partial thromboplastin time (aPTT) assays by prolonging clotting time because of Factor XII inhibition. This laboratory effect can result in misinterpretation of coagulation tests. Inform lab personnel in case the patient is receiving the treatment.
  • There is no adequate information on garadacimab‑gxii use in pregnancy, though. However, in animal studies, there was no fetal harm at high doses. Since it crosses the placenta in the third trimester, treatment should only be given if the anticipated benefits outweigh the risks.
  • Currently, it is not known if garadacimab‑gxii is excreted in breast milk. Transfer of monoclonal antibodies, like IgG, is possible, but the effects on breastfed infants are not known. Treatment should be based on the needs of the mother and the possible risks to the breastfeeding child.
  • Garadacimab-gxii has been proven safe and effective only in individuals aged 12 and above for the prevention of hereditary angioedema. Its use in children below 12 years of age is not advised because of insufficient evidence.
  • Clinical studies have examined only a small number of patients aged 65 and older. Studies indicated no significant difference in safety or efficacy in comparison with younger adults. Evaluate overall health when prescribing garadacimab-gxii 200 mg/1.2 mL injection to older patients.
