Brineura (cerliponase alfa) is a hydrolytic lysosomal N-terminal tripeptidyl peptidase indicated to slow the loss of ambulation in pediatric patients with neuronal ceroid lipofuscinosis type 2 (CLN2 disease), also known as tripeptidyl peptidase 1 (TPP1) deficiency.
Recommended Dosage:
The recommended dosage of Brineura in pediatric patients is administered once every other week by intraventricular (IV) infusion, as prescribed and supervised by your doctor. This treatment should always be administered by qualified healthcare professionals in a clinical setting. Brineura is not recommended for patients who are less than 37 weeks post-menstrual age (gestational age at birth plus post-natal age) or for patients weighing less than 2.5 kg.
If one or more doses are missed, treatment with Brineura should be restarted as soon as possible, as advised by your doctor, while maintaining the 2-week interval between infusions thereafter.
- Treatment with Brineura injection may cause serious hypersensitivity reactions, including anaphylaxis. Monitor patients closely during and after infusion. Symptoms may include rash, difficulty breathing, swelling, or low blood pressure. If severe reactions occur, immediately stop the infusion and provide appropriate medical treatment.
- Intraventricular access devices used for Brineura administration may increase the risk of meningitis or other infections. Monitor patients for vomiting, fever, headache, neck stiffness, or changes in consciousness. Prompt evaluation and treatment are necessary if infection is suspected.
