Recommended Dosage:
The recommended dosage of Lifyorli is 150 mg taken orally once on the day before, the day of, and the day after each nab-paclitaxel infusion until there is evidence of disease progression or unacceptable toxicity. Take Lifyorli with food. Swallow capsules whole. Do not crush, chew, dissolve, or split the capsules.
The recommended dosage for nab-paclitaxel is 80 mg/m2 administered as an intravenous (IV) infusion on Days 1, 8, and 15 of each 28-day cycle until there is evidence of disease progression or unacceptable toxicity.

If a dose is missed by less than 12 hours, take the missed dose. If a dose is missed by 12 hours or more, skip the missed dose and take the next dose at the regularly scheduled time. Avoid taking 2 doses at the same time to make up for a missed dose. If vomiting occurs after taking the medicine, do not take an additional dose.

Note: The recommended dosage and dosage modifications for nab-paclitaxel when administered in combination with Lifyorli differ from those for other nab-paclitaxel indications.

• Treatment with Lifyorli may reduce white blood cells (WBCs), increasing the risk of neutropenia and serious infections. Monitor blood counts regularly, watch for chills, fever, sore throat, or wounds that do not heal, and ask for immediate medical help if infection symptoms occur during treatment.
• This medicine can suppress adrenal function, causing adrenal insufficiency, especially after prolonged use or abrupt discontinuation. Taper dose gradually, monitor for dizziness, fatigue, low blood pressure, nausea, or weakness, and provide supplemental corticosteroids during stress, surgery, or serious illness periods.
• Use of Lifyorli may worsen underlying conditions typically controlled with glucocorticoids, such as arthritis, asthma, or inflammatory diseases. Assess disease stability before starting, monitor for flare symptoms, and adjust concurrent therapies appropriately to maintain control and prevent complications during the treatment course period.
• This therapeutic drug may cause fetal harm when administered during pregnancy based on its mechanism of action. Use it only if the potential benefit justifies the risk. Use effective contraception during treatment and for a defined period after stopping treatment, and counsel patients on potential risks.
• It is unknown whether this drug is excreted in human milk. Consider the benefits of breastfeeding alongside the mother’s need for therapy and potential infant risks. Patients should avoid breastfeeding during treatment and for an appropriate period after the final dose administration.
