Xgeva (denosumab) is a RANK ligand (RANKL) inhibitor used for:
- Reducing the occurrence of skeletal-related events in individuals with multiple myeloma and those with bone metastases from solid tumors.
- Treating adults and mature adolescents with a giant cell tumor of bone that cannot be surgically removed or where surgery may cause serious complications.
- Managing hypercalcemia of malignancy that doesn’t respond to bisphosphonate therapy.
Recommended Dosage:
Multiple Myeloma and Bone Metastasis from Solid Tumors: The recommended dose of Xgeva is 120 mg administered as a shot under the skin every 4 weeks in the upper arm, thigh, or belly. Your doctor may also suggest taking calcium and vitamin D supplements to prevent low calcium levels.
Giant Cell Tumor of Bone: The recommended dose of Xgeva is 120 mg once every 4 weeks. In the first month of treatment, you’ll also get additional doses of 120 mg on Days 8 and 15. It’s given as a shot in your upper arm, upper thigh, or abdomen. Your doctor may also recommend taking calcium and vitamin D supplements to prevent low calcium levels.
Hypercalcemia of Malignancy: The recommended dose of Xgeva is 120 mg administered once every 4 weeks. In the first month of treatment, you’ll also get extra doses of 120 mg on Days 8 and 15. It’s injected just beneath the skin in the upper arm, upper thigh, or abdomen.
- Xgeva and Prolia both contain the common active ingredient, denosumab. If you’re taking Xgeva, you should not take Prolia at the same time.
- Serious allergic reactions, including anaphylaxis, can happen with Xgeva 120 mg/1.7 mL. If you have a severe allergic reaction, get medical help right away and stop using Xgeva permanently.
- Xgeva may lead to severe low calcium levels in the body. Before starting Xgeva treatment, make sure any existing low calcium levels are corrected. Keep an eye on calcium levels during Xgeva therapy, especially in the beginning, and provide calcium, magnesium, and vitamin D supplements if needed.
- Some patients taking Xgeva may develop jaw problems known as osteonecrosis of the jaw (ONJ), which can cause jaw pain, infections, and tooth issues. Before starting Xgeva and during treatment, it’s important to have a dental check-up and preventive care to lower the risk of ONJ.
- Stopping denosumab treatment may lead to multiple vertebral fractures (MVF). Patients at higher risk include those with osteoporosis or previous fractures. When stopping treatment with Xgeva, assess the patient’s risk for vertebral fractures.
