Trulance (plecanatide) is a guanylate cyclase-C agonist indicated in adults for treatment of:
chronic idiopathic constipation (CIC).
irritable bowel syndrome with constipation (IBS-C).
Recommended Dosage:
The recommended dosage of Trulance for the treatment of chronic idiopathic constipation (CIC) and irritable bowel syndrome with constipation (IBS-C) is 3 mg taken orally once daily with or without food. Swallow a tablet whole for each dose.
For adult patients with swallowing difficulties, tablets can be crushed and administered orally either in applesauce or with water or administered with water via a nasogastric or gastric feeding tube. Mixing crushed tablets in other soft foods or in other liquids has not been tested.
If a dose is missed, skip the missed dose and take the next scheduled dose at the regular time. Do not take a couple of doses at the same time.
- Treatment with this therapeutic drug may cause serious dehydration in pediatric patients, especially children younger than 6 years. Serious diarrhea can cause fluid loss, low blood pressure, and hospitalization. This medicine is contraindicated in pediatric patients and should not be used in this population.
- Trulance 3 mg therapy can cause diarrhea, which may be serious in some patients. If serious diarrhea occurs, suspend dosing and rehydrate the patient. Monitor for dehydration and electrolyte imbalance, particularly in elderly patients or those with underlying medical conditions that increase risk.
- There are limited data on the use of this drug in pregnant women to inform drug-associated risks. Animal findings showed no harm at clinical doses. Use this medicine during pregnancy only if potential benefit justifies potential risk to the developing fetus and mother.
- It is unknown whether Trulance 3 mg is excreted in human milk. Because of the potential for serious side effects in breastfed infants, consider the developmental benefits of breastfeeding along with the mother’s clinical need for treatment and possible infant exposure risks.
