The drug, Nitisinone is a synthetic reversible inhibitor of 4-hydroxyphenylpyruvate dioxygenase that is used as the additional dietary restriction of Tyrosine and Phenylalanine for treating hereditary tyrosinemia type 1 (HT-1).
Recommended dosage : Healthcare professionals recommend a dose of Orfadin as 1 to 2 mg/kg divided into 2 daily doses. And the initial dose is 1 mg/kg/day, divided into morning and evening administration.

The drug, Nitisinone, is an inhibitor of 4-hydroxyphenyl-pyruvate dioxygenase, an enzyme in the tyrosine metabolic pathway. Therefore, treatment with Nitisinone may cause an increase in blood tyrosine in patients with HT-1, and patients should maintain a concomitant reduction in dietary tyrosine and phenylalanine while on Nitisinone treatment. One should maintain plasma tyrosine levels below 500 mol/L.
- Ocular signs and symptoms are reported in patients treated with Nitisinone, such as corneal ulcers, keratitis, conjunctivitis, corneal opacities, eye pain, and photophobia.
- Few patients who took Nitisinone experienced an abrupt change in neurologic status.
- Patients also experienced painful hyperkeratotic plaques on the soles and palms.
Reports show patients treated with Nitisinone and dietary restriction developed transient leucopenia, thrombocytopenia, or both. There is one case where a patient developed leukopenia and thrombocytopenia improved after the dose of Nitisinone decreased from 2 mg/kg to 1 mg/kg. One should monitor platelet, and white blood cell counts regularly during Nitisinone therapy.
