Medical Uses

KOMZIFTI (ziftomenib), from Kura Oncology and Kyowa Kirin, is indicated for the treatment of adult patients with relapsed or refractory (R/R) acute myeloid leukemia (AML) with a susceptible nucleophosmin 1 (NPM1) mutation who have no satisfactory alternative treatment options — the first and only once-daily targeted therapy for this patient population.

Ziftomenib is a menin inhibitor. It blocks the interaction between menin and the KMT2A protein complex, disrupting a key oncogenic signaling pathway that drives leukemic cell growth in NPM1-mutated AML, helping to stop the growth and spread of leukemia cells.

Warnings & Precautions

  • Differentiation syndrome, a potentially life-threatening condition, has been reported with menin inhibitors and requires prompt recognition and management.
  • QT interval prolongation is a class consideration; ECG monitoring may be required.
  • KOMZIFTI should not be started until the white blood cell (WBC) count is reduced to less than 25 x 10&sup9;/L.
  • Must be taken on an empty stomach (at least 1 hour before or 2 hours after a meal).
  • Common side effects reported in clinical trials include cytopenias and infections.
