Pimitespib is an HSP90 (heat shock protein 90) inhibitor. HSP90 is a molecular chaperone that stabilizes many oncogenic proteins, including KIT and PDGFRA — the mutant drivers of most GISTs; inhibiting HSP90 destabilizes these client proteins and suppresses tumor growth, offering an option for patients who have exhausted standard tyrosine kinase inhibitor therapy.

• Ocular disorders, including retinal disorders, have been reported; regular ophthalmologic examination is recommended.
• Diarrhea is a common and sometimes dose-limiting side effect.
• Liver function abnormalities may occur; periodic monitoring is recommended.
• Hypersensitivity reactions have been reported.
