Zejula (niraparib) is a poly (ADP-ribose) polymerase (PARP) inhibitor used:
- for the maintenance treatment of adult patients with advanced epithelial ovarian cancer, fallopian tube cancer (FTC), or primary peritoneal cancer (PPC) who are in a complete or partial response to first-line platinum-based chemotherapy.
- for the maintenance treatment of adult patients with deleterious or suspected deleterious germline BRCA-mutated recurrent epithelial ovarian cancer, fallopian tube cancer (FTC), or primary peritoneal cancer (PPC) who are in a complete or partial response to platinum-based chemotherapy.
Recommended Dosage: The recommended dosage of Zejula is taken at approximately the same time each day. Continue treatment until there is evidence of disease progression or unacceptable toxicity. First-Line Maintenance Treatment of Advanced Ovarian Cancer:
- The recommended dosage for patients weighing <77 kg (<170 lbs) OR with a platelet count of <150,000/mcL is 200 mg taken orally once daily.
- The recommended dosage for patients weighing ≥77 kg (≥170 lbs) AND who have a platelet count ≥150,000/mcL is 300 mg taken orally once daily.
- For the maintenance treatment of advanced ovarian cancer, start treatment no later than 12 weeks after their most recent platinum-containing regimen.
Maintenance Treatment of Recurrent Germline BRCA-mutated Ovarian Cancer:
- The recommended dosage is 300 mg taken orally once daily.
- For the maintenance treatment of recurrent ovarian cancer, start treatment no later than 8 weeks after their most recent platinum-containing regimen.
- In the case of a missed dose, instruct patients to take their very next dose at its regularly scheduled time. If a patient vomits or misses a dose, an additional dose of Niraparib needs to be taken.
- Treatment with Niraparib 100 mg/200 mg/300 mg tablets may increase the risk of developing myelodysplastic syndrome (MDS) or acute myeloid leukemia (AML). Monitor blood counts regularly during treatment. Seek medical advice in case unusual signs such as pale skin, fatigue, or easy bruising occur.
- This therapeutic drug can cause bone marrow suppression. This may lead to low blood cell counts. Monitor for signs and symptoms like fatigue, unusual bleeding, or infections. Regular blood tests are important to check for issues related to bone marrow.
- This medicine may cause high blood pressure and some other cardiovascular complications. Monitor blood pressure regularly. In case you experience shortness of breath, chest pain, or swelling, contact your healthcare provider (HCP) immediately.
- Posterior Reversible Encephalopathy Syndrome (PRES) can cause headaches, vision problems, confusion, and seizures. In case you experience any of these signs and symptoms, seek prompt medical help. Early detection and stopping this treatment can help prevent permanent damage.
- This therapeutic drug may cause harm to an unborn baby. Do not use it during pregnancy unless the potential benefit outweighs the risks. Women of reproductive potential should use an effective contraception method during treatment and for some time after stopping treatment with Niraparib.
- Zejula 100 mg/200 mg/300 mg tablets may pass into human breast milk and cause harm to a nursing infant. It is recommended to avoid breastfeeding while on this treatment. Consult your healthcare provider (HCP) before breastfeeding.
