Indication and Usage:

The combination medicine of sodium phenylbutyrate and taurursodiol is used to treat amyotrophic lateral sclerosis. It comes in the form of a single dose pack for oral administration in the strength of 3 g sodium phenylbutyrate & 1 g taurursodiol.

Dosage and Administration

The recommended initial dose of sodium phenylbutyrate and taurursodiol for oral suspension is 1 packet daily for the first 3 weeks. After 3 weeks, increase the maintenance dosage of 1 packet twice a day.

Preparation & Administration 

• One should empty the contents of 1 packet in a c container ontaining 8 ounces of room temperature water and stir vigorously.

Adverse Reactions

Drug Interactions

• Bile acid sequestering agents might interfere with the absorption of bile acids such as taurursodiol.
  
• Avoid the use of strong inhibitors of BSEP with this combination medicine. If concomitant use of a strong inhibitor of BSEP is deemed necessary, one should exercise caution, and monitor the serum transaminases and bilirubin is recommended.
  
• Avoid the use of aluminum-based antacids with Sodium phenylbutyrate and taurursodiol and consider other acid-lowering agents. 
  
• One should avoid the use of probenecid with Relyvrio as it might affect renal excretion of sodium phenylbutyrate metabolites.
  
• One should avoid the use of OAT1 substrates for which a small change in substrate plasma concentration might lead to serious toxicities or loss of efficacy with Relyvrio.
  
• One should avoid the concomitant use of P-gP and BCRP substrates for which a little change in substrate plasma concentration might lead to serious toxicities or loss of efficacy with Relyvrio.
  
• Professionals recommend avoiding the use of medicines that are substrates of these CYP P450 isoenzymes in which a little change in substrate plasma concentration might lead to serious toxicities or loss of efficacy.
