 Tryvio (aprocitentan) is an endothelin receptor antagonist used for the treatment of hypertension together with other antihypertensive medicines, to lower blood pressure in adult patients who are not adequately controlled by other medicines. Recommended Dosage: The recommended dosage of Tryvio is 12.5 mg orally once daily. Swallow oral tablets whole. Dosage may be taken with or without food. If a dose is missed, skip the missed dose and take the next scheduled dose at the regular time. It is not advisable to take two doses on the same day.

• The use of the medicine Tryvio 12.5 mg is contraindicated in pregnancy. To prevent pregnancy, those who can become pregnant should consider acceptable contraception before initiation of treatment, during treatment, and for one month after discontinuation of treatment.
  
• Elevations of aminotransferases and hepatotoxicity are some known effects of ERAs, including Aprocitentan. • Fluid retention and peripheral edema are some known effects of ERAs, including Aprocitentan. Monitor patients for signs and symptoms of weight gain, fluid retention, and worsening heart failure. If clinically significant fluid retention develops, treat appropriately, and discontinue treatment.
  
• Decreases in hemoglobin concentration and hematocrit may occur following the administration of ERAs including Tryvio. Measure hemoglobin before initiating therapy and periodically during treatment as clinically required.
  
• Treatment with Aprocitentan, like other ERAs, may have an adverse effect on spermatogenesis. Counsel men about potential effects on fertility.
