Zynyz (retifanlimab-dlwr) is a programmed death receptor-1 (PD-1) blocking antibody used for the treatment of adult patients with a rare and aggressive type of skin cancer called metastatic or recurrent locally advanced Merkel Cell Carcinoma (MCC).
Note: Zynyz 500 mg/20 mL (25 mg/mL) injection is approved under accelerated approval on behalf of tumor response rate as well as duration of response. Continued approval for this medication may be contingent upon verification and description of clinical benefit in confirmatory trials.
Recommended Dosage: The recommended dosage is 500 mg given as an intravenous (IV) infusion over 30 minutes every 4 weeks (28 days) until disease progression, unacceptable toxicity, or up to 24 months. Administer this medicinal product as an intravenous (IV) infusion after dilution.
Warning & Precautions
- Immune-mediated adverse reactions (IMARs), which may be serious or fatal, can occur in any organ system or tissue. Monitor patients precisely for signs and symptoms that may be clinical manifestations of underlying IMARs. Assess liver enzymes, creatinine, and thyroid function at baseline and periodically throughout treatment.
- A serious infusion-related reaction (IRR) may occur in patients receiving Zynyz injection. Monitor all the patients for signs and symptoms of IRRs. Interrupt or slow the rate of Zynyz infusion or permanently stop treatment on behalf of the severity of the reaction.
- Fatal and other critical complications can occur in those who receive allogeneic hematopoietic stem cell transplantation (HSCT) prior to or after being treated with a PD-1 or PD-L1–blocking antibody. Follow respective patients precisely for transplant-related complications and intervene immediately.
- On behalf of the mechanism of action, this medicinal product can cause fetal harm in cases used by a pregnant woman. Those with reproductive age should consider effective contraception during treatment and for 4 months following the final dose.
- Because of the probability of critical side effects in breastfed children, respective women should not breastfeed throughout treatment and for 4 months after the final dose of Retifanlimab-dlwr injection.
